Introduction
Flow cytometry is a powerful technology that enables researchers to rapidly sort live cells to conduct biological studies on purified cell populations. As these samples can contain infectious agents, precautions to protect the instrument operators and neighboring personnel from biohazards that can be generated by today's high-speed sorters are paramount.
In order to enable researchers to perform cell sorting of human cells and infectious samples, Mount Sinai School of Medicine has established a BSL2+ Cell Sorting Core located in room IMI 13-52A. In this facility, human and mouse cells with potential or known exposure to BSL-2 agents can be processed.
The BSL2+ Cell Sorting Core will not sort cells with potential or known exposure to BSL-3 agents. BSL-3 agents are transmissible through airborne routes and pose a much greater risk to users from aerosols generated during instrument use.
Equipment
The facility is equipped with a BD™ FACSAria II (SOP) cell sorter, enclosed in a Baker BioPROtect® III Hood (and installed in specially designed room with anteroom and negative pressure). The instrument accommodates five air-cooled lasers (355-nm, 405-nm, 488-nm, 561-nm and 633-nm) allowing analysis of up to 14 parameters plus FSC and SSC. The instrument is also equipment with two- and four-way bulk sorting devices for a variety of tube sizes and an automated cell deposition unit (ACDU) for sorting to multiwell plates or microscope slides. The instrument can be sterilized to perform aseptic sorts.

Reservation
The type of samples sorted have to approved by the Mount Sinai Flow Core Biosafety Committee. Reservations are made online (https://researchdb.mssm.edu/) and a biohazard assessment questionnaire must be filled out for each type of sample by clicking on Mount Sinai operations icon.

Minimum fee of one hour is charged for the use of the equipment. Sign up time is charged whether or not it is used unless a replacement user is found. Users who cancel an appointment less than 24-hours in advance are subject to the full charge of the scheduled time.
Cancelation Policy
A fee of 50 percent of the scheduled time is charged for cancellations between 24 and 72 hours prior to the scheduled time. A fee of 100 percent is charged when the cancellation occurs less than 24-hours of the scheduled time.
