The study of human embryonic stem cells (hESCs) and its differentiation into specific lineages provides an extraordinary opportunity to study the cellular / molecular mechanisms regulating pluripotency and differentiation. It also offers a potentially unlimited source of numerous cell types for drug screening and cell-based therapies. The hESC core facility has been established to facilitate the transfer of this technology to the Mount Sinai community and other NY state affiliated institutions. The objectives of this facility are:

  1. To provide undifferentiated hESCs, mouse embryonic fibroblasts (MEFs) and/or hematopoietic, cardiac and endoderm progenitors.
  2. To train faculty, postdoctoral fellows and students in the maintenance and the differentiation of hESCs.
  3. To provide tested media / reagents necessary for the maintenance as well as the differentiation of hESCs at discounted prices.
  4. To karyotype and bank hESC stocks. Currently the hESC Core maintains stocks of H1 (WA01), H7 (WA07), H9 (WA09), H13 (WA13), HES2 (ES02), HES3 (ES03) and HES4 (ES04).
  5. To generate gene targeted hESC lines (in collaboration with the mouse gene targeting facility).
  6. To assist PIs with pilot hESC projects.
  7. To assist PIs with the development of novel induced pluripotent stem (iPS) cell lines from human patients as well as their differentiation.

Induced Pluripotent Stem (iPS) Cells

Recent developments in the field have demonstrated the ability to reprogram adult human cells to an ES cell-like state using only 3 or 4 defined genes. This allows the derivation of patient-specific hiPS cell lines without the major ethical barriers associated with transfer of adult cell nuclei into oocytes. Therefore, a part of the core will be dedicated to assist investigators with the development of novel iPS cell lines and their differentiation.
